Abstract

Interleukins, a family of proteins that regulate immunity and inflammation throughout the body, are also involved in autoimmune disease (rheumatoid arthritis, multiple sclerosis, and asthma); are implicated in such other conditions as cancer, myocardial infarction, and depression; and have value in the diagnosis and prognosis of many diseases. Luke O'Neill and Andrew Bowie bring together in Interleukin Protocols a collection of standard and advanced methods for measuring these powerful agents. Their readily reproducible techniques range from the assay of interleukin protein and mRNA-using ELISA, FACS, and RT-PCR-to the study of interleukin signal transduction. Newer techniques are also covered, including the analysis of interleukin gene polymorphisms and the use of cDNA microarrays. Many of the assays are geared to specific pathologies, including breast cancer, depression, psoriasis, Grave's disease, migraine, and myocardial infarction. There are also helpful discussions of the difficulties in measuring interleukins in different biological fluids, such as sputum from asthma patients, peritoneal fluids, synovial fluid from arthritic joints, and cerbrospinal fluid from patients with meningitis. Each step-by-step method is described by a hands-on expert and includes notes on how to avoid failure. Comprehensive and highly practical, Interleukin Protocols offers biomedical investigators a stellar collection of all the major techniques needed to analyze the role of interleukins in disease, to improve diagnosis, and to foster the rapid emergence of new and more powerful therapeutics.

Abstract

More potent and selective for killing cancer cells than chemotherapy, immunotoxins constitute a new class of human therapeutics that have widespread applications and a potential that has not yet been fully recognized. In Immunotoxin Methods and Protocols, leading experts from laboratories around the world describe in step-by-step detail their cutting-edge methods for developing and applying these powerful proteins. The conjugates created by these researchers contain entire monoclonal antibodies, single chain antibody fragments, and cytokines as their carrier ligands. Their toxic portion can include such conventional chemotherapeutics as doxirubicin, such ribosome-inactivating proteins as the ricin A chain or saporin, or such bacterial toxins as diptheria toxin. Also described are a variety of applications that demonstrate the versatility of these agents, including the treatment of primary malignant brain tumors, the generation of selective brain lesions to localize critical functions, and the targeting of the AIDS virus. Each readily reproducible method is optimized for experimental success with hands-on instructions, invaluable notes on pitfalls to avoid, and informative explanations of why certain steps are necessary. Authoritative and highly practical, Immunotoxin Methods and Protocols provides today's cancer researchers, neuroscientists, and oncologists with state-of-the-art tools for exploring these powerful biological agents and their rapidly increasing applications.

Abstract

The application of recombinant DNA technology to the analysis of Epstein-Barr virus (EBV) is rapidly developing sufficient insight into the virus-host interaction, so that its role in disease pathology is now often discernible and can increasingly be interdicted. In Epstein-Barr Virus Protocols, Joanna Wilson and Gerhard May have assembled a collection of the key molecular biology protocols used in the analysis of Epstein-Barr virus, along with a series of valuable immunology, cell biology, and transgenic mouse protocols. Described in step-by-step detail by experts who use them regularly, these readily reproducible techniques include methods for gene expression with mini-EBV plasmids, for expression analysis by FISH, for EBV detection and quantitation, and for cell proliferation and death assays. In addition, the authors provide information on EBV-based vectors, an up-to-date map of EBV, a comprehensive table of available latent protein antisera, and assays from in vitro to cell to organ to organism levels. Timely and highly practical, Epstein-Barr Virus Protocols provides powerful tools for elucidating the life cycle of EBV and its host interactions, work that promises the emergence of major new treatments and cures for EBV-associated diseases, including several forms of human cancer.

Abstract

Computerized data acquisition systems are often the principle method of recording experimental results. This book takes the reader step-by-step through the process of data acquisition and analysis, explaining how to set up the systems and then obtain useful information from the data recorded. In an easy-to-read style, it guides researchers through the basics of data acquisition systems, explains the important underlying concepts, and gives numerous examples of how to analyze the recorded information. While aimed at researchers in the life sciences, the topics covered are general and will be valuable to anyone interested in learning to use data acquisition systems. The principles can be applied to the collection of data from respiratory apparatus to behavioral science experiments, and a host of other situations. Many illustrations and worked examples accompany the text, and the mathematics are kept simple. This book is an invaluable tool for the non-engineer who is collecting and analyzing experimental data using data acquisition systems.
